In particular, singletsinglet\r\nrelaxation in a two-electron-doped structure is considered. The influence of Coulomb interaction is\r\ndiscussed via comparison with a single-electron system. We find that the relaxation rate reaches similar values\r\nin the two cases but the Coulomb interaction shifts the maximum rates toward larger separations between the\r\ndots. The difference in electron-phonon interaction between deformation potential and piezoelectric coupling is\r\ninvestigated. Laterally coupled quantum dot structures doped with two electrons are considered. We take into account interaction with acoustic phonon modes via deformation potential and piezoelectric coupling. We show that piezoelectric mechanism for the considered system is of great importance and for some ranges of quantum dot molecule parameters is the dominant contribution to relaxation. It is shown that the phonon-assisted tunneling is much faster (down to ∼ 6 ps even at zero temperature) in comparison with other decoherence processes. The influence of Coulomb interaction is discussed and its consequences are indicated. We calculate the relaxation rates for GaAs quantum dot molecules and study the dependence on quantum dot size, distance and offset between the constituent quantum dots. The NQR frequency splittings, which are observed for the spin 3/2 nucleus of <jats:sup>35</jats:sup>Cl in powder samples of p-dichlorobenzene (C<jats:sub>6</jats:sub>H<jats:sub>4</jats:sub>Cl<jats:sub>2</jats:sub>) and cyanuric chloride (C<jats:sub>3</jats:sub>N<jats:sub>3</jats:sub>Cl<jats:sub>3</jats:sub>), are interpreted as a manifestation of Berry’s phase, associated with the adiabatically changing Hamiltonian due to sample rotation. The accumulation of Berry’s phase during the rotation process is responsible for the observed dependence of the NQR line shape on the rotation frequency and the asymmetry parameter. Insbesondere die Tendenz zur Ausweitung bezahlter Mitarbeit unter veränderten Bedingungen ehrenamtlichen Engagements macht den Vereinen Sorgen. Die Konsequenzen dieser Entwicklung sind noch lange nicht absehbar, und nach wie vor ist offen, wie Hauptberuflichkeit und Ehrenamtlichkeit problemlos miteinander verzahnt werden könnte. We investigated the effect of extracellular proteases on rat ENaC heterologously expressed in Xenopus laevis oocytes. Application of trypsin increased ENaC whole-oocyte currents by about 8-fold without a concomitant increase in channel surface expression. The stimulatory effect of trypsin was preserved in oocytes expressing alphagamma-ENaC, but was abolished in oocytes expressing alphabeta-ENaC. Thus, the gamma-subunit appears to be essential for channel activation by extracellular proteases. Site-directed mutagenesis of a putative prostasin cleavage site in the extracellular loop of the gamma-subunit revealed that mutating the 181Lys residue to alanine (gammaK181A) increases ENaC baseline whole-oocyte currents, decreases channel surface expression, and largely reduces the stimulatory effect of extracellular proteases (trypsin, chymotrypsin and human neutrophil elastase). In single-channel recordings from outside-out patches we demonstrated that the gammaK181A mutation essentially abolishes the activation of near-silent channels by trypsin, while a stimulatory effect of trypsin on channel gating is preserved. This apparent dual effect of trypsin on channel gating and on the recruitment of near-silent channels was confirmed by experiments using the beta518C mutant ENaC which can be converted to a channel with an open probability of nearly one by exposure to a sulfhydryl reagent. Interestingly, the gammaK181A mutation results in the spontaneous appearance of a 67 kDa fragment of the gamma-subunit in the plasma membrane which can be prevented by a furin inhibitor and also occurs after channel activation by extracellular trypsin. This suggests that the mutation promotes channel cleavage and activation by endogenous proteases. This would lower the pool of near-silent channels and explain the constitutive activation and reduced responsiveness of the mutant channel to extracellular proteases.
