Here, we investigate the adsorption of DNA origami nanostructures in\r\nnanohole arrays which represent an important class of biosensors and may benefit from the\r\nincorporation of DNA origami-based molecular probes. Nanoholes with well-defined diameter\r\nthat enable the adsorption of single DNA origami triangles are fabricated in Au films on Siwafers by nanosphere lithography. The efficiency of directed DNA origami adsorption on the\r\nexposed SiO2 areas at the bottoms of the nanoholes is evaluated in dependence of various\r\nparameters, i.e., Mg2+ and DNA origami concentrations, buffer strength, adsorption time, and\r\nnanohole diameter. We observe that the buffer strength has a surprisingly strong effect on DNA\r\norigami adsorption in the nanoholes and that multiple DNA origami triangles with 120 nm edge\r\nlength can adsorb in nanoholes as small as 120 nm in diameter. We attribute the latter\r\nobservation to the low lateral mobility of once adsorbed DNA origami on the SiO2 surface, in\r\ncombination with parasitic adsorption to the Au film. While parasitic adsorption can be\r\nsuppressed by modifying the Au film with a hydrophobic self-assembled monolayer, the limited\r\nsurface mobility of the adsorbed DNA origami still leads to poor localization accuracy in the\r\nnanoholes and results in many DNA origami crossing the boundary to the Au film even under\r\noptimized conditions. We discuss possible ways to minimize this effect by varying the\r\ncomposition of the adsorption buffer, employing different fabrication conditions, or using other\r\nsubstrate materials for nanohole array fabrication." article_type: original author: - first_name: Katharina full_name: Brassat, Katharina id: '11305' last_name: Brassat - first_name: Saminathan full_name: Ramakrishnan, Saminathan last_name: Ramakrishnan - first_name: Julius full_name: Bürger, Julius id: '46952' last_name: Bürger - first_name: Marcel full_name: Hanke, Marcel last_name: Hanke - first_name: Mahnaz full_name: Doostdar, Mahnaz last_name: Doostdar - first_name: Jörg full_name: Lindner, Jörg id: '20797' last_name: Lindner - first_name: Guido full_name: Grundmeier, Guido last_name: Grundmeier - first_name: Adrian full_name: Keller, Adrian last_name: Keller citation: ama: Brassat K, Ramakrishnan S, Bürger J, et al. Nutritional and anthropometric data at the beginning of primary school and two or four years later were available from 298 and 372 participants of IDEFICS-Germany (Identification and prevention of Dietary-induced and lifestyle-induced health Effects In Children and infants Study) and the KOPS (Kiel Obesity Prevention Study) cohort, respectively. Principal component analyses (PCA) and reduced rank regression (RRR) were used to identify dietary patterns at baseline and patterns of change in food group intake during primary school years. RRR extracted patterns explaining variations in changes in body mass index (BMI), fat mass index (FMI), and waist-to-height-ratio (WtHR). Associations between pattern adherence and excess gain in BMI, FMI, or WtHR (>75th percentile) during primary school years were examined using logistic regression. Among PCA patterns, only a change towards a more Mediterranean food choice during primary school years were associated with a favorable body composition development in IDEFICS-Germany (p < 0.05). In KOPS, RRR patterns characterized by a frequent consumption of fast foods or starchy carbohydrate foods were consistently associated with an excess gain in BMI and WtHR (all p < 0.005). In IDEFICS-Germany, excess gain in BMI, FMI, and WtHR were predicted by a frequent consumption of nuts, meat, and pizza at baseline and a decrease in the consumption frequency of protein sources and snack carbohydrates during primary school years (all p < 0.01). The study confirms an adverse impact of fast food consumption on body composition during primary school years. A. full_name: Hilbig, A. last_name: Hilbig - first_name: Lars full_name: Libuda, Lars id: '88682' last_name: Libuda orcid: 0000-0003-1603-3133 - first_name: H. full_name: Kalhoff, H. last_name: Kalhoff citation: ama: Hilbig A, Libuda L, Kalhoff H. We examined the effects of practical nutrition\r\nlessons among children with a high proportion of migration background in a\r\nprimary school setting.\r\nMethods Ten 3rd and 4th grade classes\r\n(n=166 children, 73% with migration background) received the\r\nintervention and nine school classes (n=139 children, 76%\r\nwith migration background) served as control. Before, shortly after (only\r\namong the intervention group) and three months after the three-day practical\r\nnutrition lessons, the nutrition-related skills, behavior, attitudes, and\r\nknowledge of the children were assessed using a questionnaire. Changes\r\nbetween baseline and 1st follow-up among children of the\r\nintervention group were calculated using linear mixed models. Differences\r\nbetween the two groups for changes between baseline and 2nd\r\nfollow-up were tested using linear regression analyses. Models were adjusted\r\nfor potential confounders.\r\nResults Shortly after the practical nutrition lessons, the children\r\nof the intervention group had improved their knowledge\r\n(β=1.7; 95% CI: 1.0; 2.4, P<0.001) and\r\nskills (β=1.8; 95% CI: 1.4; 2.2, P<0.001).\r\nThese changes were sustainable and larger in the intervention compared to\r\nthe control group (knowledge: β=1.6; 95% CI: 0.7;\r\n2.5, P<0.001; skills: β=1.3; 95% CI: 0.7;\r\n1.9, P<0.001). Changes in nutrition-related behavior and attitudes\r\ndid not differ between the groups.\r\nConclusions Providing practical nutrition lessons in a primary school\r\nsetting with a high proportion of children with immigrational background\r\nimproved the children’s nutrition-related knowledge and skills." author: - first_name: Katharina S. full_name: Weber, Katharina S. last_name: Weber - first_name: Jessica full_name: Eitner, Jessica last_name: Eitner - first_name: Laura full_name: Dauben, Laura last_name: Dauben - first_name: Olaf full_name: Spörkel, Olaf last_name: Spörkel - first_name: Klaus full_name: Strassburger, Klaus last_name: Strassburger - first_name: Jörn full_name: Sommer, Jörn last_name: Sommer - first_name: Barbara full_name: Kaiser, Barbara last_name: Kaiser - first_name: Anette full_name: Buyken, Anette id: '65985' last_name: Buyken - first_name: Peter full_name: Kronsbein, Peter last_name: Kronsbein - first_name: Karsten full_name: Müssig, Karsten last_name: Müssig citation: ama: Weber KS, Eitner J, Dauben L, et al. A total of 9757 three-day weighed dietary records of 1246 3- to 18-year-old participants of the Dortmund Nutritional and Anthropometric Longitudinally Designed (DONALD) study were analysed using polynomial mixed-effects regression models. Morning protein intake increased over the study period by approximately 1 % of morning energy intake (linear trend P < 0·0001), with the youngest and the oldest children having the highest protein intake (linear, quadratic trend P < 0·0001). Evening protein intake increased over time by approximately 2 % of evening energy intake in girls (linear trend P < 0·0001) and 1 % of evening energy intake in boys (quadratic trend P = 0·0313), with decreasing intake with age (girls: linear trend P < 0·0001; boys: linear trend P = 0·0963). Time trends were largely due to increases in protein from ‘starchy foods’. In conclusion, morning and evening protein intakes increased modestly between 1985 and 2014; these increases were, however, not accompanied by increases in traditional protein sources (i.e. meat or dairy products).' author: - first_name: Sarah full_name: Roßbach, Sarah last_name: Roßbach - first_name: Tanja full_name: Diederichs, Tanja last_name: Diederichs - first_name: Christian full_name: Herder, Christian last_name: Herder - first_name: Anette full_name: Buyken, Anette id: '65985' last_name: Buyken - first_name: Ute full_name: Alexy, Ute last_name: Alexy citation: ama: Roßbach S, Diederichs T, Herder C, Buyken A, Alexy U.
