At the C-terminus of each subunit, a PY-motif allows binding of the ubiquitin ligase Nedd4-2 which plays a key role in promoting ENaC retrieval from the plasma membrane. Phosphorylation of Nedd4-2 by the serum and glucocorticoid-inducible kinase 1 (Sgk1) reduces Nedd4-2 binding to the PY-motifs. In $\\beta$ and $\\gamma$ENaC, threonine residues ($\\beta$T613, $\\gamma$T623) belong to an extracellular signal-regulated kinase (ERK) motif and directly precede the PY-motifs. Thus, phosphorylation of these residues may modulate the interaction of their adjacent PY-motifs with Nedd4-2. In this study, a phosphospecific antibody was used to demonstrate phosphorylation of $\\beta$T613 in Xenopus laevis oocytes heterologously expressing rat $\\alpha$$\\beta$$\\gamma$ENaC. Treating the oocytes with progesterone to stimulate ERK increased phosphorylation of $\\beta$T613. Inactivation of the putative phosphorylation sites by mutating both threonine residues to alanine ($\\beta$T613A/$\\gamma$T623A) increased ENaC-mediated amiloride-sensitive whole-cell currents ($\\Delta$Iami) and expression of $\\beta$ENaC at the cell surface. Co-expression of Nedd4-2 largely reduced $\\Delta$Iami in oocytes expressing $\\alpha$$\\beta$$\\gamma$ENaC or channels with mutated PY-motifs in $\\alpha$ and $\\gamma$ENaC or in $\\alpha$ and $\\beta$ENaC. Importantly, the inhibitory effect of co-expressed Nedd4-2 was largely reduced in channels with mutated PY-motifs in $\\alpha$ and $\\gamma$ENaC when combined with the $\\beta$T613A mutation but conserved in channels with mutated PY-motifs in $\\alpha$ and $\\beta$ENaC combined with the $\\gamma$T623A mutation. However, less research has been done regarding proper educational design principles and guides identifying the learning-promoting characteristics as to their efficacy in an AR environment ([1], [4]). Particularly there is a big lack for design concepts in the field of preparation and accompanying tools for laboratory work, since current studies are only extending real papers or books with additional links, videos or static 3D models (e.g. [5], [6]). Hence, this paper investigates and focuses at a design concept for mobile device based AR application (App) to acquire and deepen practical skills in dealing with electro-technical laboratory equipment and components. To optimize the tooling design, a sensitivity analysis of the tool parameters that influence the quality of pipe elbows, such as mandrel height and length, is conducted. Different materials data sets including damage models were considered. Using numerical simulations, it is possible to determine an optimized tool geometry for the production of specific pipe elbow dimensions. Furthermore, as a result of the experiments and numerical simulations conducted, it is possible to increase the production velocity of the serial plant. Along with deformation, damage models are included in simulations in order to identify the right process boundaries. They can extend their lifetime at the cost of lowered performance or vice versa. This can be used to adapt failure behavior according to a maintenance plan, thus increasing availability while using up system capability fully. To facilitate setup, a control algorithm independent of a degradation model is desired. A closed loop control technique for reliability based on a health index, a measure for system degradation, is introduced. It uses self-optimization as means to implement behavior adaptation. This is based on selecting the priorities of objectives that the system pursues. Possible working points are computed beforehand using model-based multiobjective optimization techniques. The controller selects the priorities of objectives and this way balances reliability and performance. As exemplary application, an automatically actuated single plate dry clutch is introduced. The entire reliability control is setup and lifetime experiments are conducted. Results show that the variance of time to failure is reduced greatly, making the failure behavior more predictable. At the same time, the desired usable lifetime can be extended at the cost of system performance to allow for changed maintenance intervals. The membranes were conformally coated with a 5 nm thin SiO2 layer by atomic layer deposition. This layer was subsequently modified by aminopropyl silane linkers that allow further functionalization via the terminal amine group. The results evidence that in principle DNP SENS is a capable tool to analyze more complex porous systems, e.g. bioinspired functional etched ion-track membranes down to the molecular level. These results are relevant also for single nanopore systems, for which a direct analysis of the channel surface functionalization is not feasible by classical characterization methods.
