Here, we investigate the salting-out of different DNA origami shapes by the kosmotropic salt ammonium sulfate that is routinely employed in protein precipitation. We find that centrifugation in the presence of 3 M ammonium sulfate results in notable precipitation of DNA origami nanostructures but not of double-stranded genomic DNA. The precipitated DNA origami nanostructures can be resuspended in ammonium sulfate-free buffer without apparent formation of aggregates or loss of structural integrity. Even though quasi-1D six-helix bundle DNA origami are slightly less susceptible toward salting-out than more compact DNA origami triangles and 24-helix bundles, precipitation and recovery yields appear to be mostly independent of DNA origami shape and superstructure. Exploiting the specificity of ammonium sulfate salting-out for DNA origami nanostructures, we further apply this method to separate DNA origami triangles from genomic DNA fragments in a complex mixture. Our results thus demonstrate the possibility of concentrating and purifying DNA origami nanostructures by ammonium sulfate-induced salting-out.","lang":"eng"}],"user_id":"48864","publication":"International Journal of Molecular Sciences","keyword":["Inorganic Chemistry","Organic Chemistry","Physical and Theoretical Chemistry","Computer Science Applications","Spectroscopy","Molecular Biology","General Medicine","Catalysis"],"author":[{"first_name":"Marcel","full_name":"Hanke, Marcel","last_name":"Hanke"},{"full_name":"Hansen, Niklas","first_name":"Niklas","last_name":"Hansen"},{"first_name":"Ruiping","full_name":"Chen, Ruiping","last_name":"Chen"},{"last_name":"Grundmeier","full_name":"Grundmeier, Guido","first_name":"Guido"},{"last_name":"Fahmy","first_name":"Karim","full_name":"Fahmy, Karim"},{"first_name":"Adrian","full_name":"Keller, Adrian","last_name":"Keller"}],"publisher":"MDPI AG","date_created":"2022-03-07T07:28:02Z","status":"public","volume":23},{"publication":"22. But due to the lack of open and compatible simulators, authors typically create their own simulation environments for training and evaluation. This is cumbersome and time-consuming for authors and limits reproducibility and comparability, ultimately impeding progress in the field.\r\n\r\nTo this end, we propose mobile-env, a simple and open platform for training, evaluating, and comparing reinforcement learning and conventional approaches for continuous control in mobile wireless networks. mobile-env is lightweight and implements the common OpenAI Gym interface and additional wrappers, which allows connecting virtually any single-agent or multi-agent reinforcement learning framework to the environment. While mobile-env provides sensible default values and can be used out of the box, it also has many configuration options and is easy to extend. We therefore believe mobile-env to be a valuable platform for driving meaningful progress in autonomous coordination of\r\nwireless mobile networks.","lang":"eng"}],"user_id":"35343","ddc":["004"],"language":[{"iso":"eng"}],"date_updated":"2022-03-10T18:28:19Z","oa":"1","department":[{"_id":"75"}],"project":[{"_id":"1","name":"SFB 901: SFB 901"},{"_id":"4","name":"SFB 901 - C: SFB 901 - Project Area C"},{"_id":"16","name":"SFB 901 - C4: SFB 901 - Subproject C4"}],"title":"mobile-env: An Open Platform for Reinforcement Learning in Wireless Mobile Networks"},{"doi":"10.17619/UNIPB/1-1292 ","date_updated":"2022-03-11T06:08:40Z","_id":"30201","citation":{"mla":"Fanasch, Patrizia. Working life reveals challenges through technological, economic and societal development that can only rudimentarily be addressed by formal education and training. Workplace learning becomes more and more important for employees and enterprises to successfully cope with these challenges.\r\nWorkplace learning is a steadily growing field of educational research but it lacks so far a scholastic canon – there is rather a diversity of research approaches. This volume reflects this diversity by bringing together researchers from different countries and different theoretical backgrounds, presenting their current research on topics that all are relevant for understanding presages, processes and outcomes of workplace learning. The task\r\nin multiobjective optimization and multiobjective optimal control is therefore\r\nto compute the set of optimal compromises (the Pareto set) between the\r\nconflicting objectives. Since the Pareto set generally consists of an infinite\r\nnumber of solutions, the computational effort can quickly become challenging\r\nwhich is particularly problematic when the objectives are costly to evaluate as\r\nis the case for models governed by partial differential equations (PDEs). To\r\ndecrease the numerical effort to an affordable amount, surrogate models can be\r\nused to replace the expensive PDE evaluations. Existing multiobjective\r\noptimization methods using model reduction are limited either to low parameter\r\ndimensions or to few (ideally two) objectives. In this article, we present a\r\ncombination of the reduced basis model reduction method with a continuation\r\napproach using inexact gradients. The resulting approach can handle an\r\narbitrary number of objectives while yielding a significant reduction in\r\ncomputing time."}]},{"series_title":"Mathematics in Industry","language":[{"iso":"eng"}],"date_updated":"2022-03-14T07:42:01Z","doi":"10.1007/978-3-030-81455-7_23","department":[{"_id":"101"},{"_id":"655"}],"editor":[{"full_name":"Bock, H. This disruptive change in the automotive industry poses major challenges for manufacturers as well as suppliers as entirely new design and testing strategies have to be developed to remain competitive.
